Penile Erection

Penile erection is a physiological process in which the penis becomes enlarged and rigid, enabling sexual intercourse and reflecting coordinated vascular, neural, and smooth-muscle activity. Sexual stimulation activates parasympathetic signaling, promoting nitric oxide release in penile tissues; nitric oxide increases cyclic guanosine monophosphate (cGMP), which relaxes smooth muscle, expands arterial blood flow, and allows the erectile bodies to fill while compressing veins that normally drain them. Detumescence occurs when signaling declines and blood exits the erectile tissue. Studying penile erection helps explain erectile dysfunction and the effects of cardiovascular, neurological, hormonal, and metabolic disorders, while informing treatments that target blood flow and cGMP signaling.

Here, we present a protocol to offer rapid, easy, and reliable blood collection alternatives for the rat model. We describe three different blood sampling methods according to the context: tail vein puncture under anesthesia or on a conscious animal, and dorsal penile vein puncture under anesthesia.

Urethral Stricture Induction Followed by Buccal Mucosa Graft Urethroplasty in a Rat Model

In the present protocol, a urethral stricture induction was developed in Wistar rats, followed by urethral reconstruction with a buccal mucosa graft. A retrograde urethrogram and laser Doppler assessment were performed, validating urethral reconstruction (after stricture formation) and graft placement.
