Perfusion Fixation

Perfusion fixation is a tissue-preservation technique in which a fixative is circulated through an organism’s vascular system to stabilize cells and maintain tissue architecture for biological analysis. Typically, the fixative is introduced through the heart after blood is displaced, allowing solutions such as formaldehyde or paraformaldehyde to reach organs rapidly and cross-link cellular proteins. This approach produces more uniform preservation than surface immersion alone and helps limit tissue degradation, blood-related artifacts, and structural distortion. Perfusion-fixed specimens support histology, immunohistochemistry, microscopy, and neuroscience research by preserving anatomical relationships and molecular targets for detailed examination.

Presented is a method for air-inflation with vascular perfusion-fixation of the lungs that preserves the location of cells within airways, alveoli and interstitium for structure-function analyses. Constant airway pressure is maintained with an air-inflation chamber while fixative is perfused via the right ventricle. Lungs are processed for histologic studies.

The porcine model of liver normothermic machine perfusion (NMP), described here, can be successfully used to study NMP as a preservation strategy, a tool for viability assessment, and a platform for organ repair. It holds a high translational value, however it is technically challenging and labor-intensive.

Two basic types of preparation are used to visualize specimens with a light microscope: wet mounts and fixed specimens. The simplest type of preparation is the wet mount, in which the specimen is placed in a drop of liquid on the slide. A liquid specimen can be directly deposited on the slide using a dropper. Solid specimens, such as skin scraping, can be placed on the slide before adding a drop of liquid to prepare the wet mount. Sometimes the liquid is simply water, but stains are often added...
