Plant Cell Suspension

Plant cell suspension is a laboratory culture of dispersed plant cells or small cell aggregates maintained in a liquid nutrient medium, providing a controlled system for studying growth, metabolism, and cellular development. Under sterile conditions, continuous agitation keeps cells distributed and improves contact with nutrients and oxygen, while plant hormones in the medium regulate cell division, dedifferentiation, and biomass formation. These cultures support research in plant physiology, cell biology, and biotechnology, including the production of valuable secondary metabolites, evaluation of environmental or chemical responses, and propagation of plant material. Their controlled conditions also make them useful for investigating cellular processes and optimizing plant-based production systems.

Glycan Profiling of Plant Cell Wall Polymers using Microarrays

A technique called Comprehensive Microarray Polymer Profiling (CoMPP) for the characterisation of plant cell wall glycans is described. This method combines the specificity of monoclonal antibodies directed to defined glycan-epitopes with a miniature microarray analytical platform allowing screening of glycan occurrence in a broad range of biological contexts.

Targeted protein degradation represents a major regulatory mechanism for cell function. It occurs via a conserved ubiquitin-proteasome pathway, which attaches polyubiquitin chains to the target protein that then serve as molecular “tags” for the 26S proteasome. Here, we describe a simple and reliable cell-free assay for proteasomal degradation of proteins.

This protocol describes an efficient methodology for the extraction and quantification of caffeine in cell suspensions of C. arabica L. and an experimental process for evaluating the enzymatic activity of caffeine synthase with the expression level of the gene that encodes this enzyme.
