Pngase F

PNGase F is an amidase used to remove N-linked glycans from glycoproteins, making it an important tool for studying protein structure, processing, and glycosylation. The enzyme hydrolyzes the bond between the innermost N-acetylglucosamine residue and the asparagine side chain, releasing the oligosaccharide and converting asparagine to aspartic acid; it acts on most N-linked glycans but may not cleave structures with certain core fucose linkages. In biology, PNGase F treatment supports glycoprotein characterization, molecular-weight analysis, electrophoresis, immunoblotting, and mass spectrometry by simplifying complex carbohydrate-dependent patterns.

We describe a modified method for membrane fraction isolation from human cells and sample preparation for the detection of SCAP N-glycosylation and total protein by using western blot. We further introduce a GFP-labeling method to monitor SCAP trafficking using confocal microscopy. This protocol can be used in regular biology laboratories.

A Quantitative Glycomics and Proteomics Combined Purification Strategy

A high-throughput protocol was developed for combined proteomics and glycomics purification and LC-MS/MS quantification in plasma. Deamidation analysis of N-linked glycosylation motifs was specific to deglycosylated sites. Accurate quantitation of N-glycans was achieved by coupling filter aided N-glycan separation to the individuality normalization when labeling with glycan hydrazide tags strategy.
