Primary Sympathetic Neurons

Primary sympathetic neurons are autonomic nerve cells isolated directly from sympathetic ganglia and maintained in culture to study how the sympathetic nervous system develops and functions. They respond to stimulation by generating action potentials, conducting signals along axons, and releasing neurotransmitters such as norepinephrine at target tissues. In laboratory models, these neurons support investigations of neuronal survival, differentiation, axon growth, synapse formation, and responses to growth factors or toxic compounds. Their use provides physiologically relevant insight into autonomic regulation and helps researchers examine mechanisms underlying neural development, injury, drug effects, and disorders involving sympathetic signaling.

This video demonstrates an in vitro method to generate postganglionic sympathetic neurons from human pluripotent stem cells (hPSCs). hPSCs are differentiated into neural crest cells (NCCs) on a basement membrane matrix support using a differentiation medium. The NCCs are then dissociated and formed into spheroids using another differentiation medium. Finally, the spheroids are transformed into postganglionic sympathetic neurons using a sympathetic neuron medium.
