Recombinant Protein Mutagenesis

Recombinant protein mutagenesis is the deliberate alteration of a gene encoding a recombinant protein to examine or improve the protein’s structure, function, stability, or production. Researchers introduce specific changes into a DNA sequence, express the modified gene in a host system such as bacteria or cultured cells, and compare the resulting protein with the original through purification and functional assays. In biology, this approach links amino acid sequence to properties such as binding, catalytic activity, folding, and solubility. It supports enzyme engineering, antibody optimization, disease-mechanism studies, and the development of proteins for biotechnology and therapeutic research.

Mutagenesis and Functional Selection Protocols for Directed Evolution of Proteins in E. coli

Here we demonstrate a simple protocol to create a random mutant library for a given target sequence. We show how this method, which is performed in vivo in Escherichia coli, can be coupled with functional selections to evolve new enzymatic activities.

A Protocol for Functional Assessment of Whole-Protein Saturation Mutagenesis Libraries Utilizing High-Throughput Sequencing

We present a protocol for the functional assessment of comprehensive single-site saturation mutagenesis libraries of proteins utilizing high-throughput sequencing. Importantly, this approach uses orthogonal primer pairs to multiplex library construction and sequencing. Representative results using TEM-1 β-lactamase selected at a clinically relevant dosage of ampicillin are provided.
