Reference Electrode Preparation

Reference electrode preparation is the process of producing a stable electrochemical reference with a known, reproducible potential for measuring voltage in biological and chemical systems. Typically, a silver/silver chloride element contacts a defined chloride-containing electrolyte, such as potassium chloride, while a porous junction permits ionic conduction and limits mixing with the sample. Careful preparation includes cleaning the electrode, maintaining electrolyte concentration, removing air bubbles, and checking the junction for blockage or leakage. Well-prepared reference electrodes improve the accuracy and stability of measurements in electrophysiology, electrochemistry, biosensors, and studies of cellular or tissue responses.

In this article, we present a cell design that accommodates multiple reference electrodes, along with the assembly steps to assure maximum accuracy of the data obtained in electrochemical measurements.

Neural Activity Propagation in an Unfolded Hippocampal Preparation with a Penetrating Micro-electrode Array

We have developed an in vitro unfolded hippocampus which preserves CA1-CA3 array of neurons. Combined with the penetrating micro-electrode array, neural activity can be monitored in both the longitudinal and transverse orientations. This method provides advantages over hippocampal slice preparations as the propagation in the entire hippocampus can be recorded simultaneously.
