Rho Zero Cells

Rho zero cells are cells that lack mitochondrial DNA, providing a powerful model for studying how mitochondrial genes support cellular function. Because mitochondrial DNA encodes essential components of the respiratory chain, its loss disrupts oxidative phosphorylation and ATP production, often forcing cells to rely more heavily on glycolysis for energy. Researchers use Rho zero cells to separate mitochondrial and nuclear genetic effects, investigate mitochondrial-nuclear communication, and examine mechanisms of respiratory deficiency. These cells also help clarify how mitochondrial dysfunction contributes to disease and provide a controlled system for testing genetic complementation, metabolic adaptation, and potential treatments targeting mitochondrial disorders.

Affinity Precipitation of Active Rho-GEFs Using a GST-tagged Mutant Rho Protein (GST-RhoA(G17A)) from Epithelial Cell Lysates

The method presented here describes an assay to follow activation of RhoA specific GDP/GTP Exchange Factors (GEFs) in cultured cells by making use of a mutant RhoA GST fusion protein that has high affinity for activated GEFs. GEFs are precipitated from cell lysates, detected by Western blotting and quantified by densitometry.
