Sds-insoluble Proteins

SDS-insoluble proteins are proteins that remain in an undissolved fraction after treatment with sodium dodecyl sulfate (SDS), a detergent commonly used to disrupt protein structure and confer a negative charge. Their resistance to solubilization can result from extensive aggregation, strong hydrophobic interactions, covalent crosslinking, or association with other cellular materials, preventing complete dispersion in the detergent solution. Studying this fraction helps researchers distinguish soluble and insoluble protein populations during cell extraction and biochemical analysis. SDS-insoluble proteins are also relevant to protein misfolding, inclusion bodies, and disease-associated aggregates, while specialized solubilization methods can support their characterization by electrophoresis and mass spectrometry.

Testing protein-protein interaction is indispensable for dissection of protein functionality. Here, we introduce an in vitro protein-protein binding assay to probe a membrane-immobilized protein with a soluble protein. This assay provides a reliable method to test interaction between an insoluble protein and a protein in solution.
