Sirna Delivery

siRNA delivery is the process of transporting small interfering RNA molecules into target cells so they can selectively reduce gene expression, making it an important tool in molecular biology and therapeutic research. After entering the cytoplasm, siRNA is incorporated into the RNA-induced silencing complex, whose guide strand directs sequence-specific recognition and cleavage of complementary messenger RNA. Delivery systems such as lipid nanoparticles, polymers, and conjugated molecules help protect siRNA from degradation and improve cellular uptake, tissue distribution, and release. These approaches support functional gene studies and the development of treatments for diseases driven by aberrant or harmful gene activity.

The goal of this protocol is to use cationic/anionic liposomes with a neuro-targeting peptide as a CNS delivery system to enable siRNA to cross the BBB. The optimization of a delivery system for treatments, like siRNA, would allow for more treatment options for prion and other neurodegenerative diseases.
