Spasticity

Spasticity is a motor disorder characterized by a velocity-dependent increase in muscle tone that limits normal movement and often follows damage to upper motor neurons. When a muscle is stretched rapidly, reduced descending inhibition can heighten spinal stretch-reflex activity, causing involuntary contraction, stiffness, and exaggerated resistance to movement. Spasticity commonly occurs after stroke, spinal cord injury, cerebral palsy, or multiple sclerosis, and its distribution and severity can affect posture, walking, and daily activities. Understanding its neural mechanisms supports clinical assessment, rehabilitation planning, and treatments designed to reduce abnormal tone while preserving useful muscle function.

Children with spastic cerebral palsy (SCP) have limb spasticity, movement disorders, and abnormal posture due to injury to the cerebral cortex motor area, resulting in inability to stand and walk normally. Therefore, alleviating limb spasticity and enhancing gross motor function in children with SCP have become important therapeutic goals.

Using a robotic isokinetic device with electromyography (EMG) measurements, this protocol illustrates that isokinetic motion itself can improve inter-rater reliability for the angle of catch measurements in stroke patients with mild elbow flexor spasticity.

Impaired mitochondrial transport and morphology are involved in various neurodegenerative diseases. The presented protocol uses induced pluripotent stem cell-derived forebrain neurons to assess mitochondrial transport and morphology in hereditary spastic paraplegia. This protocol allows characterization of mitochondrial trafficking along axons and analysis of their morphology, which will facilitate the study of neurodegenerative disease.
