Spectrophotometer Readout

A spectrophotometer readout is the quantitative measurement produced when an instrument assesses how a sample interacts with light, typically by reporting absorbance or transmittance at a selected wavelength. The instrument compares light passing through the sample with a reference, and absorbance values can reflect the concentration of light-absorbing molecules according to the Beer-Lambert law. In biology, researchers use these measurements to estimate nucleic acid or protein concentrations, monitor enzyme activity, and evaluate cell or biochemical assays. Reliable interpretation depends on appropriate blanks, calibration, wavelength selection, and measurements within the instrument’s linear range.

Simple Bulk Readout of Digital Nucleic Acid Quantification Assays

We describe an endpoint digital assay for quantifying nucleic acids with a simplified (analog) readout. We measure bulk fluorescence of droplet-based digital assays using a standard qPCR machine rather than specialized instrumentation and confirm our results by microscopy.

A Molecular Readout of Long-term Olfactory Adaptation in C. elegans

Here we describe a molecular readout of long-term olfactory adaptation in Caenorhabditis elegans. The Protein Kinase G, EGL-4, is necessary for stable adaptation responses in the primary sensory neuron pair called AWC. During prolonged odor exposure EGL-4 translocates from the cytosol to nucleus of the AWC.

Microvolume Protein Concentration Determination using the NanoDrop 2000c Spectrophotometer

Microvolume samples are quantified by a spectrophotometer system that uses natural surface tension to retain samples without the use of cuvettes or capillaries. The dynamic range of protein concentrations and speed by which they can be measured are greatly increased with this method.
