Stem Cuttings

Stem cuttings are a vegetative propagation method in which a section of stem produces new roots and shoots, allowing a plant to reproduce without seeds. After a cutting is removed from a parent plant, it can form adventitious roots at the cut or along the stem when moisture, oxygen, temperature, and other environmental conditions support tissue survival and growth. This method preserves the parent plant’s genetic characteristics and is widely used to multiply ornamental, crop, and research plants. Studying stem cuttings also helps biologists examine plant regeneration, tissue differentiation, and the factors that regulate organ formation.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
