Trap Stiffness Calibration

Trap stiffness calibration is the process of determining how strongly an optical tweezers system holds a microscopic particle, a critical step for converting particle displacement into quantitative force measurements in biological research. Calibration typically relates the bead’s thermal fluctuations to the restoring force generated by the optical trap, using methods such as the equipartition theorem or analysis of the bead’s Brownian-motion power spectrum. Accurate stiffness values allow researchers to measure forces exerted by molecular motors, protein interactions, and cells, supporting studies of mechanobiology, intracellular transport, and the physical properties of biomolecules.

Here, a method that enables quick, efficient, and inexpensive preparation of polyacrylamide gels in a multiwell plate format is described. The method does not require any specialized equipment and could be easily adopted by any research laboratory. It would be particularly useful in research focused on understanding stiffness-dependent cell responses.
