Tris Elution

Tris elution is a laboratory method that uses a Tris-based buffer to recover biological molecules, especially DNA, from a solid matrix such as an agarose gel or binding membrane. The buffer maintains a stable, near-neutral pH while hydration, diffusion, and changes in molecular interactions release the target molecule into solution; subsequent centrifugation or collection separates the eluate from the supporting material. In biology, this approach helps purify DNA fragments after electrophoresis for cloning, sequencing, amplification, or other downstream analyses. Its effectiveness depends on buffer composition, contact time, temperature, and the properties of the matrix and target molecule.

A Novel Method to Determine the Longitudinal Antibacterial Activity of Drug-Eluting Materials

Here, we present a protocol to evaluate the antibacterial efficacy of an antibiotic-eluting polymer to simulate prophylactic clinical application by using a commercially available real-time ATP-based luminescent microbial viability assay. This method enables the monitoring of the longitudinal activity of drug-eluting materials and can be widely adapted to test anti-microbial drug delivery platforms.

Gallium(III) 5,10,15-(tris)pentafluorophenylcorrole and its freebase analogue exhibit low micromolar cell cytotoxicity. This manuscript describes an RNA transcription reaction, imaging RNA with an ethidium bromide-stained gel, and quantifying RNA with UV-Vis spectroscopy, in order to assess transcription inhibition by corroles and demonstrates a straightforward method of evaluating anticancer candidate properties.
