Virus Separation

Virus separation is the isolation of virus particles from cells, tissues, culture fluids, or other biological materials, enabling their analysis and controlled use in biology. It exploits differences in particle size, density, charge, or surface properties through methods such as filtration, centrifugation, and chromatography, often followed by purification and concentration steps. These approaches help researchers distinguish viruses from host-cell components, assess viral structure and infectivity, prepare samples for molecular and imaging studies, and support vaccine, diagnostic, and antiviral research. Careful separation improves sample quality and reproducibility while preserving the biological properties needed for downstream experiments.

Here, we describe a single cell micro-aspiration method for the separation of infected amoebae. In order to separate viral subpopulations in Vermamoeba vermiformis infected by Faustoviruses and unknown giant viruses, we developed the protocol detailed below and demonstrated its ability to separate two low-abundance novel giant viruses.
