Wild-type Host

A wild-type host is an organism that carries the naturally occurring or standard genetic background used as a reference in biological research. Because it lacks a specific engineered or experimentally introduced mutation, its cells, tissues, and physiological responses provide a baseline for comparison with mutant, transgenic, or otherwise modified hosts. Researchers use wild-type hosts to assess gene function, host-pathogen interactions, infection outcomes, and responses to environmental or experimental conditions. Comparing these results with those from altered hosts helps identify how particular genes or biological pathways influence development, disease susceptibility, immunity, and other organismal traits.

Wild-Type Blocking PCR to Detect Low-Frequency Somatic Mutations

This video describes wild-type blocking polymerase chain reaction, or WTB-PCR, which detects somatic mutations. The PCR-based technique uses a locked nucleic acid, or LNA, an oligonucleotide that binds to its complementary wild-type allele and blocks its elongation. The blocking selectively allows the amplification of low-level mutant alleles over their wild-type variants in the sample.

We provide a cost-effective and rapid molecular genotyping protocol that employs variety-specific PCR primers that target DNA sequence differences within the chloroplast trnL-F spacer region to differentiate between varieties of Imperata cylindrica (cogongrass) that cannot be distinguished by morphology alone. These varieties include the federally listed noxious weed, cogongrass and closely-related, wide-spread ornamental variety, I. cylindrica var. koenigii (Japanese blood grass).
