Cytokine Release Measurement

Cytokine release measurement is the quantitative assessment of signaling proteins secreted by immune, tumor, or engineered cells, providing a readout of cellular activation and inflammatory communication. In cancer research, samples such as cell-culture supernatants or blood are analyzed with immunoassays, including ELISA, multiplex bead-based assays, or ELISpot, in which cytokine-specific antibodies capture and detect targets through enzyme, fluorescence, or spot-based signals. Measuring factors such as interferons, interleukins, and tumor necrosis factor helps characterize immune responses, evaluate immunotherapies and cell therapies, monitor treatment-related inflammation, and compare disease models. Standardized sampling and normalization improve the reliability of these measurements.

Accurate and Simple Measurement of the Pro-inflammatory Cytokine IL-1β using a Whole Blood Stimulation Assay

We describe a simple immunoassay to measure the production of pro-inflammatory cytokines, such as IL-1 beta production, in patients presenting with autoinflammatory phenotypes. By activating cells in whole blood cultures with pathogen-associated molecular patterns, specifically with lipopolysaccharide, cytokine secretion can be conveniently evaluated in whole blood supernatants.

Described here is a method for producing exogenous cytokine in patient-derived xenograft (PDX) mice via weekly intraperitoneal injection of a cytokine-transduced stromal cell line. This method broadens the utility of PDX and provides the option for transient or sustained exogenous cytokine delivery in a multitude of PDX models.

Single Cell Measurement of Dopamine Release with Simultaneous Voltage-clamp and Amperometry

The amperometric technique measures dopamine release from a single cell by detecting the oxidative current produced by spontaneous dopamine oxidization. Simultaneous voltage clamp and amperometry methodology reveal the mechanistic relationship between the overall "activity" of dopamine transporter and the regulatory role of this activity on the reverse transport of dopamine.
