Diffuse Midline Glioma

Diffuse midline glioma is an aggressive, infiltrative central nervous system tumor that arises in midline structures such as the brainstem, thalamus, or spinal cord and remains a major challenge in cancer research. Many tumors harbor alterations affecting histone H3, particularly H3 K27-altered variants, which disrupt chromatin regulation and gene expression to promote abnormal cell growth and invasion. Diagnosis integrates neuroimaging, tissue examination when feasible, and molecular profiling, while radiation therapy provides temporary disease control for some patients. Research focuses on understanding tumor biology, developing targeted and immune-based treatments, and improving clinical trial strategies for this difficult-to-treat cancer.

Glial cells isolated from tumor tissue via an enzymatic digestion are cultured in a neurobasal medium, which promotes their growth and the development of free-floating neurospheres.

IDH1-mutant diffuse low-grade gliomas are rare tumors that are difficult to culture. We introduce a simple, non-enzymatic explant method that preserves IDH1-mutant protein expression and sustains cultures for months. Importantly, this technique works with cryopreserved tumors, offering a valuable resource for glioma research.
