Mda-mb231 Cells

MDA-MB-231 cells are a human breast cancer cell line widely used as an in vitro model of aggressive, triple-negative breast cancer, making them valuable for studying tumor biology. In culture, these adherent cells grow under controlled nutrient, temperature, and gas conditions, while assays can measure their viability, migration, invasion, gene expression, and treatment responses; their receptor profile lacks estrogen receptor, progesterone receptor, and HER2 expression. Researchers use MDA-MB-231 cells to investigate metastatic behavior, cancer cell signaling, drug sensitivity, and interactions with the tumor microenvironment. Results from this model inform preclinical cancer research but require validation in additional models and patient-derived systems.

Invasive Behavior of Human Breast Cancer Cells in Embryonic Zebrafish

Here, we describe xenograft zebrafish models using two different injection sites, i.e., perivitelline space and duct of Cuvier, to investigate the invasive behavior and to assess the intravasation and extravasation potential of human breast cancer cells, respectively.

This method utilizes zebrafish embryos to efficiently test the vascular invasive ability of cancer cells. Fluorescent cancer cells are injected into the precardiac sinus or yolk sac of developing embryos. Cancer cell vascular invasion and extravasation is assessed via fluorescence microscopy of the tail region 24 to 96 hr later.

The cytotoxic activity of the phenanthridine PJ-34 in cancer cells undergoing mitosis was documented in real time by live confocal imaging. PJ-34 eradicated human breast cancer MDA-MB-231 cells harboring extra-centrosomes in mitosis. Unlike normal bi-focal mitosis, the extra-centrosomes were not clustered in the two spindle poles in the presence of PJ-34.
