Mouse Kidney Isolation

Mouse kidney isolation is a laboratory procedure that removes one or both kidneys from a mouse for structural, cellular, molecular, or functional analysis. The process involves humane euthanasia, careful dissection through the abdominal cavity, separation of the kidneys from surrounding tissues, and preservation in an appropriate medium for downstream processing. Isolated kidneys can be examined by histology, immunostaining, gene or protein analysis, cell dissociation, or organ culture. In cancer research, this tissue supports studies of renal tumors, tumor-associated changes, therapeutic responses, and interactions between malignant cells and the kidney microenvironment.

In this report we describe a method for the isolation and culture of the progenitor cell niche from the embryonic mouse kidney that can be used to study signaling pathways regulating stem/progenitor cells of the developing kidney. These cultured cells are highly accessible to small molecule and recombinant protein treatment, and importantly also to viral transduction, which allows efficient manipulation of candidate pathways.

DNabT cells are rare among peripheral T cells; however, they are abundant in certain non-lymphoid tissues. Difficulty of isolating DN T cells from non-lymphoid tissue hinders their functional analysis despite increasing recognized pathophysiologic significance. We describe a novel protocol for isolation of highly purified DN T cells from murine kidney.
