Nadh Oxidative Phosphorylation

NADH oxidative phosphorylation is the mitochondrial process that converts reducing energy in NADH into ATP, the main energy currency of cells. NADH donates electrons to respiratory Complex I, initiating electron transport through the inner mitochondrial membrane; the resulting proton gradient drives ATP synthase to produce ATP while oxygen serves as the terminal electron acceptor. In cancer research, this pathway helps explain how tumor cells balance mitochondrial respiration, glycolysis, biosynthesis, and redox control during growth or metabolic stress. Measuring or disrupting NADH oxidation and oxidative phosphorylation can reveal cancer-specific metabolic dependencies and inform studies of therapeutic response.

The objective is to monitor the mitochondrial redox state of isolated hearts within the context of physiologic preload and afterload pressures. A biventricular working rabbit heart model is presented. High spatiotemporal resolution fluorescence imaging of NADH is used to monitor the mitochondrial redox state of epicardial tissue.

Here we describe a method to directly visualize microregional tissue hypoxia in the mouse cortex in vivo. It is based on concurrent two-photon imaging of nicotinamide adenine dinucleotide (NADH) and the cortical microcirculation. This method is useful for high resolution analysis of tissue oxygen supply.
