Nitrosamine Standard Synthesis

Nitrosamine standard synthesis is the preparation of well-characterized N-nitrosamine reference compounds used to identify and measure these potentially carcinogenic contaminants in biological, pharmaceutical, and environmental samples. The process typically involves controlled nitrosation of a secondary amine, followed by isolation, purification, and analytical characterization to confirm the compound’s identity and concentration. In cancer research, these standards support LC-MS and related assays that quantify nitrosamines and their metabolites, helping investigators evaluate exposure, metabolism, DNA-damaging potential, and cancer risk. Reliable standards also improve method validation, comparison across studies, and the development of strategies to detect and reduce nitrosamine formation.

2° Amines to N-Nitrosamines: Reaction with NaNO2

2023

Secondary amines react with nitrous acid to form N-nitrosamines, as depicted in Figure 1. Nitrous acid, a weak and unstable acid, is formed in situ from an aqueous solution of sodium nitrite and strong acids, such as hydrochloric acid or sulfuric acid, in cold conditions. In the presence of an acid, the nitrous acid gets protonated. The subsequent loss of water results in the formation of the electrophile known as nitrosonium ion. Figure 1. The nitration reaction of secondary amines The...

Synthesis of a Deuterated Standard for the Quantification of 2-Arachidonoylglycerol in Caenorhabditis elegans

2019

This work describes a robust and straightforward method to detect and quantify the endocannabinoid 2-arachidonoylglycerol (2-AG) in C. elegans. An analytical deuterated standard us prepared and used for the quantification of 2-AG by isotopic dilution and liquid chromatography-electrospray ionization-tandem mass spectrometry (LC-ESI-MS/MS).

Sparks, Department of Materials Science and Engineering, The University of Utah, Salt Lake City, UT Hydrogels are a versatile class of cross-linked polymers produced through relatively simple procedures and with generally inexpensive materials. They can be formed from solution and involve a polymer backbone formed from monomer reagents, an initiator which makes the polymer reactive and a crosslinking species which binds the polymer chains...

Source: Vy M. Dong and Diane Le, Department of Chemistry, University of California, Irvine, CA Merrifield's solid-phase synthesis is a Nobel Prize winning invention where a reactant molecule is bound on a solid support and undergoes successive chemical reactions to form a desired compound. When the molecules are bound to a solid support, excess reagents and byproducts can be removed by washing away the impurities, while the target compound remains bound to the resin. Specifically, we will...

Synthesis and Decomposition Reactions

2020

Synthesis and decomposition are two types of redox reactions. Synthesis means to make something, whereas decomposition means to break something. The reactions are accompanied by chemical and energy changes. Synthesis Reactions Synthesis reactions are also called combination reactions. It is a reaction in which two or more substances combine to form a complex substance. Synthesis reactions are generally represented as: A + B → AB or A + B → C. The formation of nitrogen dioxide is a synthesis...
