Pma Ionomycin

PMA ionomycin is a laboratory stimulation treatment that combines phorbol 12-myristate 13-acetate (PMA) with the calcium ionophore ionomycin to induce strong, receptor-independent cellular signaling. PMA activates protein kinase C by mimicking diacylglycerol, while ionomycin transports calcium across cellular membranes, elevating intracellular Ca2+ and activating calcium-dependent pathways; together, these signals can drive transcription, cytokine production, and other activation responses. In cancer research, PMA ionomycin is used to probe signaling networks, stimulate immune cells for functional assays, and assess how tumors or candidate treatments influence immune activation, providing a controlled reference for studying inflammation, immunosuppression, and therapeutic response.

This protocol is based on LPS and ATP-induced death of PMA-differentiated THP-1 macrophages. We use flow cytometry to analyze Annexin V and 7-AAD double staining to detect cell death, using the whole cell and employing scanning electron microscopy to observe the cell membrane morphology.

An efficient protocol for the ex vivo expansion of tumor-reactive T cells from tumor-draining lymph nodes or other secondary lymphoid tissues of tumor-bearing hosts is described. This protocol selectively expands tumor-specific T cells for use in adoptive immunotherapy of breast cancer.

This video demonstrates the in vitro differentiation of granulocyte-macrophage-colony-stimulating factor or GM-CSF-producing T helper cells (THGM). Isolated naïve T cells are stimulated for differentiation into THGM, which is confirmed by detecting high intracellular GM-CSF production and a low expression of other T helper cell-specific cytokines.

T-lymphocyte mitogenesis is accompanied by blastogenic transformation, whereupon the cell volume enlarges before cell division. Here, we describe a method to quantify blastogenesis in T lymphocytes using an automated cell counter with the capability of measuring cell diameters.
