Femur Tibia Isolation

Femur tibia isolation is a laboratory dissection technique used to recover the femur and tibia, commonly as sources of bone marrow and skeletal tissue for research. The procedure involves exposing the hindlimb, removing surrounding muscle and connective tissue, separating the bones at their joints, and preserving the marrow by opening the bone ends for collection. In cancer research, isolated bones support the study of marrow-derived cells, hematopoiesis, tumor cell colonization, and interactions within the bone microenvironment. Samples can subsequently undergo cell culture, flow cytometry, histological analysis, or molecular assays to investigate disease mechanisms and evaluate potential treatments.

Murine Hind Limb Long Bone Dissection and Bone Marrow Isolation

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.

One constraint of preclinical research in the field of bone repair is the lack of experimental control over the local mechanical environment within a healing bone lesion. We report the design and use of an external fixator for bone repair with the ability to change fixator stiffness in vivo.
