Primary Cancer Cells

Primary cancer cells are tumor-derived cells isolated directly from a patient or an animal model, providing a biologically relevant system for studying cancer beyond established cell lines. Researchers obtain these cells from tumor tissue, dissociate the sample, and maintain the resulting population under controlled culture conditions that support survival while preserving features such as genetic diversity, cellular heterogeneity, and tumor-specific behavior. In cancer research, primary cancer cells help investigate disease mechanisms, drug responses, treatment resistance, and interactions with the tumor microenvironment. Their closer representation of tumors can improve the relevance of preclinical findings, although limited growth, variability between samples, and demanding culture requirements can complicate their use.

Pancreatic cancer stem cells (CSCs) can be expanded in vitro using the anchorage-independent sphere culture technique, which represents a powerful tool to study CSC biology and can serve as the first step to develop novel CSC-targeting therapies. Here the methodology for expanding, analyzing and targeting of pancreatic CSCs is provided.
