Molecular Precursors

Molecular precursors are molecules or chemical substances that serve as starting materials for producing other molecules, making them central to planning and controlling chemical synthesis. Through reactions such as bond formation, cleavage, oxidation, reduction, or functional-group interconversion, a precursor changes under defined conditions into a desired product or an intermediate. Chemists select precursors based on molecular structure, reactivity, availability, and compatibility with solvents, catalysts, and temperature. In organic, materials, and pharmaceutical chemistry, precursor design supports the preparation of complex compounds, enables efficient synthetic routes, and helps researchers connect molecular transformations with properties and practical applications.

Source: Zhao, K., et al. An In Vitro Model to Study the Effect of 5-Aminolevulinic Acid-mediated Photodynamic Therapy on Staphylococcus aureus Biofilm. J. Vis. Exp. (2018)The video demonstrates a photodynamic therapy protocol to assess antibacterial effects on Gram-positive bacterial biofilms. Biofilms are divided into four groups: two receive a photosensitizer precursor, and two receive a buffer. All are incubated in the dark, followed by red light exposure to one precursor-treated group and...

Stimulation of Notch Signaling in Mouse Osteoclast Precursors

2017

Notch signaling is a form of cellular communication that relies upon direct contact between cells. To properly induce Notch signaling in vitro, Notch ligands must be presented to cells in an immobilized state. This protocol describes methods for in vitro stimulation of Notch signaling in mouse osteoclast precursors.

We describe a small molecule-based protocol for differentiation of mouse embryonic stem cells into oligodendrocyte precursor cells (OPCs). This protocol generates Olig2+NG2+ OPCs with high efficiency by 30 days of differentiation. We also describe a method to generate "spiking" OPCs that can fire action potentials.

(2018)This video demonstrates the transplantation of interneuron precursor cells into the neonatal mouse brain using either a homotopic or heterotopic approach. The procedure involves securing an anesthetized recipient pup, identifying the target brain region, and injecting a suspension of precursor cells derived from a donor pup at the same developmental stage.

Molecular Orbital (MO) Theory

Source: Tamara M. Powers, Department of Chemistry, Texas A&M University This protocol serves as a guide in the synthesis of two metal complexes featuring the ligand 1,1'-bis(diphenylphosphino)ferrocene (dppf): M(dppf)Cl2, where M = Ni or Pd. While both of these transition metal complexes are 4-coordinate, they exhibit different geometries at the metal center. Using molecular orbital (MO) theory in conjunction with 1H NMR and Evans method, we will determine the geometry of these two...
