Activity-dependent Growth

Activity-dependent growth is the process by which cells, tissues, or neural circuits change their growth and maturation in response to functional activity. During development, patterns of electrical signaling, muscle use, or sensory input can alter intracellular calcium levels and activate gene expression, protein synthesis, and cytoskeletal remodeling, thereby strengthening selected connections or shaping tissue structure. This feedback between activity and development helps refine neural circuits, coordinate organ formation, and match biological architecture to functional demands. Studying activity-dependent growth clarifies how physiological signals influence development and can inform research on neurodevelopmental disorders, regeneration, and recovery after injury.

Here we describe a procedure allowing a detailed analysis of the phosphorylation-dependent activation of the IRF3 transcription factor. This is achieved through the combination of a high resolution SDS-PAGE and a native-PAGE coupled to immunoblots using multiple phosphospecific antibodies.

Detection of the pH-dependent Activity of Escherichia coli Chaperone HdeB In Vitro and In Vivo

2016

This study describes biophysical, biochemical and molecular techniques to characterize the chaperone activity of Escherichia coli HdeB under acidic pH conditions. These methods have been successfully applied for other acid-protective chaperones such as HdeA and can be modified to work for other chaperones and stress conditions.

Here we describe biochemical assays that can be used to characterize ATP-dependent chromatin remodeling enzymes for their abilities to 1) catalyze ATP-dependent nucleosome sliding, 2) engage with nucleosome substrates, and 3) hydrolyze ATP in a nucleosome- or DNA-dependent manner.

Chemical Kinetics The reaction rate is the speed at which a chemical reaction occurs. The reaction rate is defined as the change in concentration of a component in the reaction with time. The speed of a reaction depends on several factors, including the concentration of reactants and the temperature at which the reaction is performed. Each reactant contributes to the speed of the reaction by a specific factor. This relationship is defined by the reaction rate law. Rate Law The rate law is an...

Chemical Kinetics and the Reaction Rate Law Chemical kinetics refers to the rate or speed of a chemical reaction. The rate depends on the mechanism, complexity, and number of reactants in the reaction. Reactant concentration also plays a significant role in the rate of a reaction. The rate law quantifies this relationship through experimentation. Each reactant contributes to the speed of the reaction by a factor known as the reaction order. This factor can range from zero to two, and it depends...
