Whole-mount Microscopy

Whole-mount microscopy is an imaging approach that visualizes an entire small organism, tissue, embryo, or organ without physically slicing it into sections, preserving spatial relationships central to developmental biology. Samples are typically fixed, permeabilized, and stained with antibodies, dyes, or reporter signals, then optically sectioned through their depth using confocal or light-sheet microscopy to reconstruct three-dimensional patterns. This method reveals cell distributions, tissue morphogenesis, lineage markers, and gene-expression domains across developing specimens. By linking molecular signals with anatomy, whole-mount microscopy supports studies of embryonic patterning, organ formation, developmental defects, and tissue organization.

Light Sheet Microscopy Imaging and Mounting Strategies for Early Zebrafish Embryos

A sample preparation strategy for imaging early zebrafish embryos within an intact chorion using a light-sheet microscope is described. It analyzes the different orientations that embryos acquire within the chorion at the 70% epiboly and bud stages and details imaging strategies for obtaining cellular-scale resolution throughout the embryo using the light-sheet system.

Here, we describe a protocol for whole mount immunofluorescence and image-based quantitative volumetric analysis of early stage mouse embryos. We present this technique as a powerful approach to qualitatively and quantitatively assess cardiac structures during development, and propose that it may be widely adaptable to other organ systems.
