Antibody Evaluation

Antibody evaluation is the systematic assessment of an antibody’s binding, specificity, affinity, and biological activity to determine whether it is suitable for research or clinical use. It typically measures recognition of a target antigen through methods such as enzyme-linked immunosorbent assay, immunoblotting, flow cytometry, or surface-based binding assays, while functional tests examine effects such as neutralization or receptor blocking. In immunology and infection research, these analyses help validate diagnostic antibodies, characterize immune responses, compare vaccine-induced or infection-induced antibodies, and identify candidates for therapeutic development. Rigorous evaluation also reveals cross-reactivity, variability, and assay limitations that can affect experimental interpretation.

An In Vitro Assay to Evaluate the Effect of Antibodies on Candida tropicalis Biofilm

This video demonstrates an in vitro technique to study the effect of antibodies on fungal biofilms. The pathogenic fungus Candida tropicalis secretes a virulence factor secreted aspartyl proteinase 2 (Sap2), facilitating biofilm formation. Upon introducing a heat-inactivated serum containing Sap2-specific antibodies that inhibit biofilm maturation, the viability of the fungal cells is assessed using a chromogenic assay.

Scalable High Throughput Selection From Phage-displayed Synthetic Antibody Libraries

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.

Utilizing common methods to evaluate peptide conjugation, AC and payload intracellular accumulation, and tumor targeting, this protocol helps researchers during the key initial development phases.
