Bhk T7-9 Cells

BHK T7-9 cells are a baby hamster kidney-derived cell line that constitutively expresses bacteriophage T7 RNA polymerase, making them a valuable platform for studying viral gene expression and infection. Inside these cells, T7 RNA polymerase recognizes T7 promoter sequences on introduced plasmids and transcribes viral or other target RNAs, supporting protein production or, for suitable systems, recovery of recombinant viruses. In immunology and infection research, BHK T7-9 cells help investigate viral replication, host responses, and pathogen-host interactions, while enabling reverse-genetics experiments that modify defined viral genes to examine their effects on infectivity, immune evasion, and disease-related phenotypes.

Source: Bouillier, C. et. al., Amplification, and Titration of Recombinant Respiratory Syncytial Viruses. J. Vis. Exp. (2019)This video demonstrates the generation of a green fluorescent protein (GFP)-expressing recombinant respiratory syncytial virus using a T7-based reverse genetics system. The co-transfection of plasmids encoding viral proteins and a GFP-tagged genome leads to protein expression and initiates virus rescue.
