Calcium-induced Signaling

Calcium-induced signaling is a cellular communication process in which changes in intracellular calcium concentrations regulate physiological responses. Signals such as receptor activation or membrane depolarization can open calcium channels, allowing Ca2+ to enter the cytoplasm or triggering its release from intracellular stores; calcium-binding proteins then translate these changes into altered enzyme activity, gene expression, secretion, or cytoskeletal behavior. In immunology and infection, this pathway helps control lymphocyte activation, phagocytosis, cytokine production, and antimicrobial responses. Pathogens can also manipulate calcium dynamics to alter host-cell functions, making calcium signaling important for understanding immune regulation, infectious disease mechanisms, and potential therapeutic targets.

We developed a gene-chimeric preparation of ventral hippocampal – accumbens circuit in vitro that allows direct live imaging to analyze presynaptic mechanisms of nicotinic acetylcholine receptors (nAChRs) mediated synaptic transmission. This preparation also provides an informative approach to study the pre- and post-synaptic mechanisms of synaptic plasticity.

Assessing T-cell Receptor-Induced Calcium Influx Using a Calcium Indicator Dye

This video analyzes T-cell receptor-induced calcium influx in T-cells. Anti-CD3 antibodies activate the T-cell signaling pathways, leading to an increase in intracellular calcium ions that bind with Indo-1 dye. In flow cytometry, a spectrum shift from the calcium-free state to the calcium-bound state correlates with T-cell receptor-induced calcium influx.

Intercellular Ca2+-waves are driven by gap junction channels and hemichannels. Here, we describe a method to measure intercellular Ca2+-waves in cell monolayers in response to a local single-cell mechanical stimulus and its application to investigate the properties and regulation of gap junction channels and hemichannels.

Real-time In Vivo Recording of Arabidopsis Calcium Signals During Insect Feeding Using a Fluorescent Biosensor

This protocol outlines a simple method for analyzing calcium signals in plants generated by feeding hemipteran insects, such as aphids. Arabidopsis thaliana transformed with the GFP calcium biosensor GCaMP3 allow for the real-time in vivo imaging of calcium dynamics with a high temporal and spatial resolution.
