Intracranial Injection

Intracranial injection is a specialized technique for delivering a defined substance directly into the brain or a surrounding compartment, enabling controlled investigation of central nervous system biology and disease. Typically performed under anesthesia with stereotactic guidance, the method uses anatomical coordinates and a fine needle to administer a measured inoculum while minimizing damage to adjacent tissue. In immunology and infection research, intracranial injection can model pathogen entry into the CNS, examine neuroinvasion and tissue tropism, and characterize local immune responses that may be difficult to study after peripheral exposure. The approach also supports evaluation of antimicrobial interventions and mechanisms of infection-associated inflammation.

Source: Liou, C. et al. Intracerebroventricular Delivery of Gut-Derived Microbial Metabolites in Freely Moving Mice. J. Vis. Exp. (2022).This video demonstrates the intracranial injection of short-chain fatty acids (SCFAs) in a freely moving mouse to study their effects on brain-mediated behavior. An injector cannula is inserted into a guide cannula implanted in the mouse brain. After recovery, the mouse acclimates in a novel cage. SCFAs are then administered, and locomotor behavior is recorded.

Modeling Brain Metastases Through Intracranial Injection and Magnetic Resonance Imaging

Intracranial brain metastasis modeling is complicated by an inability to monitor tumor size and response to treatment with precise and timely methods. The presented methodology couples intracranial tumor injection with magnetic resonance imaging analysis, which when combined, cultivates precise and consistent injections, enhanced animal monitoring, and accurate tumor volume measurements.

Synergistic Effect of Intracranial and Intraperitoneal Delivery of Analgesic Drugs on Thermal Pain Response in a Rat

Source: Martinez, E., et al. Intracranial Pharmacotherapy and Pain Assays in Rodents. J. Vis. Exp. (2019).This video demonstrates the intracranial administration of an analgesic drug in an anesthetized rat with a pre-implanted guide cannula in the prefrontal cortex. An additional analgesic drug is co-administered intraperitoneally. Following recovery, the rat’s thermal pain response is assessed using Hargreave’s apparatus, where paw withdrawal latency is measured to evaluate the analgesic...
