Nadph Oxidase

NADPH oxidase is an enzyme complex that transfers electrons from NADPH to molecular oxygen, generating reactive oxygen species (ROS) in cells. In immune cells such as neutrophils and macrophages, activation assembles membrane and cytosolic subunits, enabling the respiratory burst that produces superoxide and downstream oxidants within phagosomes. These reactive molecules help destroy engulfed bacteria and fungi, making NADPH oxidase central to innate immune defense and infection control. Defects in the complex can cause chronic granulomatous disease, while excessive or misregulated ROS production may contribute to tissue injury and inflammatory disease.

NADPH oxidase is the major source of reactive oxygen species (ROS) in phagocytes. Because of the ephemeral nature of ROS, it is difficult to measure and monitor ROS levels in living animals. A minimally invasive method for serial quantification of ROS in living mice is described.

This is a straightforward histochemical technique and is useful in investigating mitochondrial diseases, aging, and aging-related disorders.
