Neutrophil Phagocytosis

Neutrophil phagocytosis is an innate immune process in which neutrophils recognize, engulf, and destroy invading microorganisms and damaged material, providing rapid protection during infection. Surface receptors bind microbial molecules directly or through opsonins such as antibodies and complement, triggering actin-driven engulfment into a phagosome. The phagosome then fuses with granules, whose antimicrobial enzymes and reactive oxygen species help kill and digest the captured target. Studying this process helps explain host defense, inflammation, and susceptibility to infection, while phagocytosis assays support research into neutrophil function, immune deficiencies, pathogen survival strategies, and therapies that influence inflammatory responses.

This manuscript describes a simple method to measure the phagosomal pH and area as well as the cytoplasmic pH of human and mouse neutrophils using the ratiometric indicator seminaphthorhodafluor (SNARF)-1, or S-1. This is achieved using live-cell confocal fluorescence microscopy and image analysis.
