Nycodenz Gradient Separation

Nycodenz Gradient Separation is a density-gradient centrifugation technique that isolates cells, organelles, or microorganisms according to their buoyant density, making it useful for obtaining defined populations from complex biological samples. Nycodenz, a nonionic iodinated medium, is prepared at selected concentrations to form a density gradient; during centrifugation, particles migrate until they reach a position where their density matches that of the surrounding solution. In immunology and infection research, the method can enrich leukocytes, mononuclear cells, or other sample-specific fractions for downstream phenotyping, culture, microscopy, or molecular analysis. By reducing sample complexity, gradient separation supports more consistent characterization of host immune responses and pathogen-associated material.

This protocol describes enzymatic digestion of mouse skin in nutrient-rich medium followed by gradient separation to isolate leukocytes. Cells thus derived can be used for diverse downstream applications. This is an effective, economical, and improved alternative to tissue dissociation machines and harsher trypsin and dispase-based tissue digestion protocols.

We demonstrate the use of discontinuous density gradients to separate bacterial populations based on capsule production. This method is used to compare capsule amount between cultures, isolate mutants with a specific capsule phenotype, or to identify capsule regulators. Described here is the optimization and running of the assay.
