Renal Subcapsular Space

The renal subcapsular space is the narrow potential space between the kidney’s fibrous capsule and the outer renal cortex, forming a confined interface between protective tissue and functioning parenchyma. During subcapsular injection, a needle separates the capsule from the cortex to create a localized site where introduced cells, tissue grafts, antigens, or biomaterials can remain in contact with nearby renal tissue and receive nutrients from surrounding vessels. In immunology and infection research, this access route supports studies of graft rejection, inflammatory responses, immune-cell activity, and host reactions to implanted or infectious material. Its defined anatomy also enables controlled comparisons of local versus systemic effects.

We provide a simple and efficient method to transplant 2’-deoxyguanosine treated E18.5 thymus into the renal capsule of a nude mouse. This method should aide in the study of both thymic epithelial cells function and T cells maturation.
