Treg Function Measurement

Treg function measurement is the assessment of how effectively regulatory T cells suppress immune responses, providing insight into immune tolerance, inflammation, and host defense. In a typical suppression assay, purified Tregs are co-cultured with activated responder T cells, and their activity is quantified by measuring responder-cell proliferation, cytokine production, or activation markers, often alongside phenotypic markers such as FOXP3 and CD25. These measurements help distinguish Treg abundance from functional capacity and support studies of infection, autoimmunity, transplantation, and immunotherapy. Standardized functional readouts can also clarify how pathogens or treatments alter immune regulation and inform therapeutic development.

A functional assessment of the neuromuscular junction (NMJ) can provide essential information on the communication between muscle and nerve. Here we describe a protocol to comprehensively evaluate both the NMJ and muscle functionality using two different muscle-nerve preparations, i.e. soleus-sciatic and diaphragm-phrenic.

Functional Reconstitution and Channel Activity Measurements of Purified Wildtype and Mutant CFTR Protein

Described here is a rapid and effective procedure for functional reconstitution of purified wild-type and mutant CFTR protein that preserves activity for this chloride channel, which is defective in Cystic Fibrosis. Iodide efflux from reconstituted proteoliposomes mediated by CFTR allows studies of channel activity and the effects of small molecules.

Measuring Respiratory Function in Mice Using Unrestrained Whole-body Plethysmography

The assessment of respiratory physiology has traditionally relied upon techniques, which require restraint or sedation of the animal. Unrestrained whole-body plethysmography, however, provides precise, non-invasive, quantitative analysis of respiratory physiology in animal models. In addition, the technique allows repeated respiratory assessment of mice allowing for longitudinal studies.
