Vesicular Stomatitis Virus

Vesicular stomatitis virus (VSV) is an enveloped, negative-sense RNA virus in the Rhabdoviridae family that causes vesicular disease primarily in livestock and serves as an important model for viral immunology. Its bullet-shaped virion uses the surface glycoprotein to enter host cells, while the viral RNA-dependent RNA polymerase transcribes and replicates the genome in the cytoplasm. The matrix protein can suppress host gene expression, enabling rapid viral production while shaping innate immune responses such as interferon signaling. VSV supports studies of antiviral defense, viral pathogenesis, vaccine and viral-vector design, and oncolytic therapy, making it valuable in infection and translational research.

We have developed a simple and reproducible protocol to access stomatal response to live bacteria. This method minimizes wounding and manipulation of the leaf as compared to the use of epidermal peels reported previously.

Unraveling how physiology and morphology are linked allows a deeper understanding of mechanistic functioning of plant leaves. We present both a procedure to derive parameters of stomatal regulation from stomatal conductance measurements and correlations with traditional functional leaf traits.

Ex Vivo Infection of Live Tissue with Oncolytic Viruses

Oncolytic viruses are promising for cancer therapeutics. The ability to ascertain the infectability of live tissue specimens obtained from patients prior to treatment is a unique advantage of this therapeutic approach. This protocol describes how to process tissues for ex vivo infection with oncolytic virus and subsequent viral quantification.
