Muscle Contractile Function

Muscle contractile function is the ability of muscle tissue to generate force and movement, a fundamental process in normal physiology and clinical medicine. In skeletal muscle, motor neuron signals trigger calcium release from the sarcoplasmic reticulum; calcium binds troponin, shifts tropomyosin, and permits myosin heads to pull actin filaments through sarcomeres using ATP. The resulting contraction can produce shortening, tension, or controlled resistance to lengthening. Assessing contractile function helps researchers and clinicians understand weakness, fatigue, neuromuscular disorders, cardiac dysfunction, and muscle injury, while informing diagnostic testing, rehabilitation strategies, and the development of therapies that target excitation-contraction coupling or force production.

Ex Vivo Assessment of Contractility, Fatigability and Alternans in Isolated Skeletal Muscles

We describe a method to directly measure muscle force, muscle power, contractile kinetics and fatigability of isolated skeletal muscles in an in vitro system using field stimulation. Valuable information on Ca2+ handling properties and contractile machinery of the muscle can be obtained using different stimulating protocols.

Presented here is a protocol to assess the contractile properties of striated muscle myofibrils with nano-Newton resolution. The protocol employs a setup with an interferometry-based, optical force probe. This setup generates data with a high signal-to-noise ratio and enables the assessment of the contractile kinetics of myofibrils.
