Brain Derived Neurotrophic Factor Supplementation

Brain-derived neurotrophic factor (BDNF) supplementation is the experimental delivery of BDNF to cells, tissues, or organisms to assess how this neurotrophin affects neuronal development, survival, and plasticity. BDNF binds tropomyosin receptor kinase B (TrkB), activating intracellular signaling pathways that regulate synaptic function, neurite growth, and resistance to cellular stress. In neuroscience research, supplementation helps clarify BDNF’s role in learning, memory, neurodevelopment, and repair after injury. These studies can also inform investigations of neurodegenerative and psychiatric disorders, while highlighting the challenges of delivering biologically active BDNF to the relevant nervous-system targets.

Primary, human fetal brain-derived, multipotential progenitor cells proliferate in vitro while maintaining the capacity to differentiate into neurons and astrocytes. This work shows that neural progenitors can be induced to differentiate through stages of the oligodendrocytic lineage by conditioning with select growth factors.

Here we provide a reproducible method to examine adult neurogenesis using a neurosphere assay derived from the whole brain or from either the telencephalic, tectal or cerebellar regions of the adult zebrafish brain. Additionally, we describe the procedure to manipulate gene expression in zebrafish neurospheres.
