Fitc Ferritin Permeation

FITC-ferritin permeation is a tracer-based method that uses fluorescein isothiocyanate-labeled ferritin to assess how macromolecules cross biological barriers, including the blood-brain barrier. After introduction into the circulation or experimental tissue, the fluorescent ferritin provides a detectable signal whose distribution indicates barrier passage, endothelial uptake, or transport through vesicular pathways. In neuroscience, this approach helps evaluate blood-brain barrier integrity, vascular permeability, and alterations caused by injury, inflammation, disease, or experimental treatments. Fluorescence imaging of tissue sections can reveal where the tracer accumulates, supporting studies of neurovascular function and the mechanisms that regulate molecular exchange between blood and brain.

This video demonstrates a method to study permeation through an in vitro blood-brain barrier (BBB) model using astrocytes and endothelial cells on a membrane insert, with free FITC and FITC-loaded ferritin. FITC-ferritin bound to receptors on endothelial cells and crossed the membrane barrier to the lower chamber while free FITC showed restricted permeation and remained in the upper chamber.

A method to establish an in vitro model of blood-brain barrier based on a co-culture of rat brain microvascular endothelial cells and astrocytes is described and validated. This system proved to be a valid tool to study the effect of nanoformulation on the trans-barrier permeation of fluorescent molecules.
