Lead Citrate Staining

Lead citrate staining is an electron microscopy method that increases contrast in ultrathin biological sections by depositing electron-dense lead salts. In neuroscience, tissue sections are typically treated with lead citrate after fixation and resin embedding, allowing the reagent to associate with anionic cellular structures and scatter electrons more strongly than surrounding material. This contrast reveals neuronal membranes, synaptic vesicles, mitochondria, myelin, and other organelles in transmission electron micrographs, supporting analysis of synaptic organization, axonal pathology, and ultrastructural changes in neurodegenerative or experimental models.

This video demonstrates procedures for characterization of human pancreatic islets using hematoxylin and eosin (H&E) and immunohistochemistry (IHC). Pancreatic sections from head, body, and tail regions are stained by both H&E and IHC to determine islet endocrine composition (insulin, glucagon, and pancreatic polypeptide), cell replication (Ki67), and inflammatory infiltrates (H&E, CD3). The uncinate region is localized using IHC for pancreatic polypeptide.
