Microdissection Brain Regions

Microdissection of brain regions is a technique for isolating anatomically defined areas from brain tissue so their cells, molecules, or structural features can be studied separately. Researchers use anatomical landmarks and specialized cutting tools to remove small tissue samples while preserving regional identity, often from freshly collected, fixed, or sectioned specimens. The isolated material supports analyses of gene expression, proteins, cellular composition, and pathology, helping link molecular changes to specific neural circuits and functions. In neuroscience, brain-region microdissection strengthens comparisons across areas, developmental stages, disease models, and treatment conditions, providing more precise insight than analyses of whole-brain tissue.

We present a hands-on, step-by-step, rapid protocol for mouse brain removal and dissection of discrete regions from fresh brain tissue. Obtaining brain regions for molecular analysis has become routine in many neuroscience labs. These brain regions are immediately frozen to obtain high quality transcriptomic data for system level analysis.

Non-Laser Capture Microscopy Approach for the Microdissection of Discrete Mouse Brain Regions for Total RNA Isolation and Downstream Next-Generation Sequencing and Gene Expression Profiling

RNA expression profiling of discrete mouse brain regions requires a precise and repeatable tissue collection strategy. A protocol that uses both coronal brain sectioning and tissue corer-assisted microdissection is described here. The yield and quality of total RNA obtained from the resulting samples confirms the utility of the outlined method.
