Morphological Analysis

Morphological analysis is the systematic study of an organism, cell, or tissue’s form and structural features, providing a foundation for understanding how anatomy relates to function. In neuroscience, it examines neurons and neural circuits by visualizing features such as soma shape, dendritic branching, axon organization, and synaptic architecture through microscopy, staining, and image-based measurements. Researchers use these structural data to classify cell types, compare developmental or disease-related changes, and relate neuronal organization to connectivity and activity. Morphological analysis supports studies of brain development, neurodegeneration, injury, and circuit organization, while quantitative approaches improve the reproducibility of cellular and tissue-level comparisons.

Automated Sholl Analysis of Digitized Neuronal Morphology at Multiple Scales

We have developed a computer program to analyze neuronal morphology. In combination with two existing open source analysis tools, our program performs Sholl analysis and determines the number of neurites, branch points, and neurite tips. The analyses are performed so that local changes in neurite morphology can be observed.

We present a protocol that permits to view and to quantitatively asses the morphology of the dendritic tree of individual Purkinje cells grown in organotypic cerebellar slice cultures. This protocol is intended to promote studies on the mechanisms of Purkinje cell dendritic development.
