Peripheral Sensory Nerve Fibers

Peripheral sensory nerve fibers are extensions of primary afferent neurons that detect conditions outside the central nervous system and transmit sensory information to the spinal cord and brain. Specialized peripheral endings convert mechanical, thermal, or chemical stimuli into receptor potentials, which trigger action potentials that travel along myelinated A fibers or unmyelinated C fibers through peripheral nerves and dorsal root ganglia. These fibers support touch, temperature, pain, and proprioception, while differences in conduction speed and receptor specialization shape sensory perception. Understanding their organization and signaling is essential for studying neural circuits, sensory disorders, nerve injury, chronic pain, and potential therapies for neuropathy.

This video demonstrates the staining technique of resin-embedded, osmium tetroxide-treated rat peripheral nerve sections using toluidine blue. The stained nerve sections help visualize the fine structures of peripheral nerves.

Tissue Preparation and Immunostaining of Mouse Sensory Nerve Fibers Innervating Skin and Limb Bones

Immunocytochemical identification of peripheral sensory nerve fiber subtypes (and detection of protein expression therein) are key to the understanding of molecular mechanisms underlying peripheral sensation. Here we describe methods for preparation of peripheral/visceral tissue samples, such as skin and limb bones, for specific immunostaining of peripheral sensory nerve fibers.

In Vivo Single-Fiber Recording of Sciatic Nerve C-Fibers in Rats

2025

This video demonstrates a single-fiber recording of C-fiber nerve conduction in rats. An anesthetized rat is incised to expose the sciatic nerve, which provides sensory and motor supply to the lower limbs. The individual C-fibers are then exposed. Electrical stimulation is provided using stimulus electrodes implanted in the foot, and a response is recorded from a single nerve fiber.

(2020).This video demonstrates the surgical construction of a composite regenerative peripheral nerve interface (C-RPNI) in a rat model of peripheral nerve injury. The procedure involves attaching a transected peroneal nerve to a muscle graft and overlaying it with a dermal skin graft to promote nerve regeneration. Over time, motor and sensory neurons...

Visualization of Mitochondria within Intraepidermal Nerve Fibers

Source: Hamid, H. S., et al. Three-dimensional Imaging and Analysis of Mitochondria within Human Intraepidermal Nerve Fibers. J. Vis. Exp. (2017) This video demonstrates the method for visualizing nerve-specific mitochondria using three-dimensional imaging. It outlines the process of acquiring sequential scans of pre-labeled intraepidermal nerve fibers and generating 3D surface images to isolate and display mitochondria in relation to nerve structures.
