Recurrent Tumor Therapy

Recurrent tumor therapy refers to the treatment of cancer that returns after an initial response or period of remission, a major challenge in neuro-oncology because tumors may infiltrate sensitive brain tissue and develop resistance. Management begins with imaging and, when feasible, tissue or molecular analysis to distinguish recurrence from treatment-related changes and guide therapy. Depending on tumor location, biology, prior treatment, and patient condition, clinicians may combine repeat surgery, focused or external-beam radiation, systemic drugs, targeted therapies, or clinical trials. These approaches aim to control tumor growth, preserve neurological function, relieve symptoms, and extend survival while informing research on resistance and personalized care.

Tumor Treating Field Therapy in Combination with Bevacizumab for the Treatment of Recurrent Glioblastoma

A novel methodology that is employed for the treatment of recurrent glioblastomas is described. This treatment approach employs the application of alternating electric tumor treating fields (TTFields), known as TTF Therapy in combination with bevacizumab, a targeted agent that is currently FDA approved as monotherapy.

Luciferase-modified human brain tumor xenografts can be established intracranially in athymic mice, with subsequent monitoring of tumor growth and response to therapy using bioluminescence imaging. In combination with survival analysis, bioluminescence monitoring is an essential research tool for pre-clinical testing of therapies being considered for treating brain tumors.

Many types of human brain tumors are localized to specific regions within the brain and are difficult to grow in culture. This protocol addresses the role of tumor microenvironment and investigates new drug treatments by analyzing fluorescent primary brain tumor cells growing in an organotypic mouse brain slice.
