Rpe Functional Assays

RPE functional assays are laboratory tests that evaluate how retinal pigment epithelial cells perform the specialized activities required to support retinal health. These assays typically measure properties such as epithelial barrier formation, nutrient and waste transport, secretory signaling, and phagocytosis of shed photoreceptor outer segments under controlled culture conditions. In neuroscience research, functional readouts help distinguish mature, healthy RPE cells from dysfunctional or diseased cells and complement structural analyses of retinal models. They support studies of retinal degeneration, drug responses, cell-based therapies, and tissue engineering by linking cellular behavior to photoreceptor maintenance and visual-system function.

2017

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.

In this video we describe a technique to isolate iris and retinal pigment epithelial cells from porcine eyes to obtain their primary cultures for in vivo and ex vivo studies. These pigment cells could be genetically modified to study regenerative approaches to treat ocular disorders.

Extraction of Tissue Antigens for Functional Assays
