Semi Thin Microtome

A semi-thin microtome is an instrument used to cut thin sections from resin-embedded tissue for microscopic examination, bridging routine histology and ultrastructural analysis. In neuroscience, tissue is typically fixed, dehydrated, embedded in epoxy or acrylic resin, and sectioned as the block advances in precisely controlled increments against a glass or diamond knife. These sections preserve cellular organization for light microscopy of brain and peripheral nerve samples, helping researchers assess neuronal layers, myelin, axonal pathology, and lesion boundaries before selecting regions for transmission electron microscopy.

Semi-automated Optical Heartbeat Analysis of Small Hearts

We have developed a Semi-automated Optical Heartbeat Analysis method (SOHA) for analyzing high speed optical recordings from Drosophila, zebrafish and embryonic mouse hearts. We demonstrate the application of our methodology to the analysis of heart function in fruit fly and embryonic mouse hearts.

A Semi-quantitative Approach to Assess Biofilm Formation Using Wrinkled Colony Development

We provide a simple, semi-quantitative method to investigate biofilm formation in vitro. This method takes advantage of the Zeiss stemi 2000-C Dissecting Microscope (with camera attachment) to monitor both the timing and pattern of biofilm formation, as assessed by the development of wrinkled colonies.

Analysis of vestibular hair cell function is complicated by their location deep within the hardest part of the skull, the petrous temporal bone. Most functional hair cell studies have used acutely isolated hair cells. Here we describe a semi-intact preparation of mouse vestibular epithelium for electrophysiological and two-photon microscopy studies.

Whole blood Immunophenotyping is indispensable for monitoring the human immune system. However, the number of reagents required and the instability of specific fluorochromes in premixed cocktails necessitates daily reagent preparation. Here we show that semi-automated preparation of staining antibody cocktail helps establish reliable immunophenotyping by minimizing variability in reagent dispensing.
