Septum Isolation

Septum isolation is an experimental neuroscience procedure that separates the brain’s septal region from surrounding tissue and neural pathways to clarify its distinct functions. Researchers use targeted dissection or surgical transection to disconnect septal nuclei from selected inputs and outputs while preserving the tissue of interest for anatomical, physiological, or behavioral analysis. This approach helps reveal how septal circuits contribute to limbic network activity, motivation, emotional behavior, learning, and memory. By reducing the influence of neighboring structures, septum isolation provides a controlled framework for linking specific neural connections with observable functions and for interpreting broader brain-circuit organization.

Here, we describe an improvement of the semi-in vitro (SIV) method for observing pollen tube guidance and reception in Arabidopsis thaliana, which increases the receptivity of ovules. The high-throughput SIV cum septum method may be coupled with gametophyte marker lines and genetically encoded biosensors to monitor the dynamic process of fertilization.

We describe here a method for biopsying olfactory mucosa from rat and human nasal cavities. These biopsies can be used for either identifying molecular anomalies in brain diseases or isolating multipotent adult stem cells that can be utilized for cell transplantation in animal models of brain trauma/disease.
